The Bray Bomber showed her amateur pedigree and style throughout the ten-round contest, drawing Serrano in, making her miss and punishing her on the counter.
She promised the best performance of her career and certainly delivered one of them as she did her talking in the ring, just as she said she would.
However, there was a nervy moment when the first judge's decision of a 95-95 draw was called out, but the other two had it 97-93 in her favour.
It takes Taylor's record to 3-0 against her great rival as she defended her super-lightweight world titles and undisputed status at Madison Square Garden in New York.
The victory adds to her wins over Serrano at MSG in April 2022 and on the Jake Paul v Mike Tyson card last November in Texas. The first of those was by majority decision, while the other was unanimously at the AT&T Stadium.
Taylor, who turned 39 last year, will now have a decision to make regarding if she wants to fight on.
The bout headlined an all-female card at the Mecca of Boxing, with 17 world titles on the line across the card.
Earlier in the night, Chantelle Cameron defeated Canadian Jessica Camara by unanimous decision to retain her WBC super-lightweight interim world title.
Cameron, the only person to have beaten Taylor in the professional ranks, warmed to the task against Camara in the third fight of the night.
The Northampton native had a challenging week due to a visa issue and was stuck in the UK while the problem was resolved.
She only arrived in America 48 hours before stepping in the ring and missed Thursday's press conference as a result.
Fighting under trainer Stephen 'Swifty' Smith for the first time, Cameron showed her usual relentlessness, using her jab well throughout the ten-round bout and causing significant damage to Camara's left eye, which was looked at by the doctor ahead of the eighth round.
All three judges scored the bout massively in the 34-year-old's favour: 99-91, 98-92 and 99-91
Cameron landed 215 punches to her opponent's 142, having thrown 552 to Camara's 486.
